Eric, did you get AVR Studio working with your STK500?

I had 2 problems:

1) If your laptop goes to sleep, when it wakes up the STK500 driver is flaked out.

Simply unplug the USB cable and plug it back in

 

2) I had the 10 pin cable plugged in backwards.
